This vent integrates a hydrophobic, low-extractable ePTFE membrane with a weldable or mechanically sealable carrier, designed for direct integration into the gas inlet and exhaust ports of shake flasks, benchtop fermenters, seed train bioreactors, and large production-scale microbial and cell culture vessels. It provides a sterile, bacteria-retentive airway that allows the massive volumes of oxygen required for aerobic microbial growth to enter the vessel, and the equally large volumes of carbon dioxide to exit, while maintaining an absolute sterile barrier that prevents the contamination of the culture. The vent's hydrophobic property protects against the wetting and clogging caused by foam, condensate, and media splashes, ensuring consistent gas exchange throughout the batch.

Cause

Aerobic microbial fermentation—whether producing recombinant proteins, antibiotics, vaccines, or industrial enzymes—requires a continuous supply of sterile oxygen. A large-scale E. coli or yeast fermentation can consume oxygen at rates requiring vessel aeration of 1-2 vessel volumes per minute. The air must be sterile; a single bacterial or fungal contaminant entering the bioreactor can outcompete the production organism, destroying a batch worth hundreds of thousands to millions of dollars. Fermentation broth foams heavily, and this foam, carrying media components and microorganisms, can rise into the exhaust vent and clog a non-hydrophobic filter. Similarly, condensate from the warm, humid exhaust air can wet a standard filter membrane, causing it to become air-blocked. The vent must maintain its high airflow and sterility throughout the batch—which may last from hours to weeks—despite foam, moisture, and aggressive cleaning and sterilization-in-place (CIP/SIP) cycles.

The ePTFE sterile vent provides a reliable, high-capacity sterile gas exchange pathway. The hydrophobic ePTFE membrane is inherently resistant to wetting by water, foam, and cell culture media. Foam that contacts the membrane collapses and drains back, rather than wetting and clogging the pores. The 0.2-micron pore size provides sterilizing-grade bacterial retention, ensuring that all air entering the vessel is sterile and all air exiting is contained. The membrane's high porosity and thin profile provide the very low pressure drop needed for high-volume aeration. The vent carrier can be welded or clamped into a stainless steel or single-use plastic bioreactor port. The membrane withstands repeated steam sterilization (SIP at 134°C) and is compatible with gamma-sterilized single-use bioreactor systems.

How It Works

The vent is installed on the bioreactor's air inlet and exhaust ports. Sterile compressed air is pushed through the inlet vent into the vessel. The vent's 0.2-micron membrane provides a final sterilizing filter, ensuring that any contaminants from the air supply line are captured. As the culture respires, CO₂-rich exhaust air exits through the exhaust vent to the atmosphere or a waste gas system. The hydrophobic membrane prevents foam and condensate from clogging the exhaust path. At the end of the batch, the vessel and vents are cleaned and steam-sterilized in place, or the single-use system is disposed of.
